腦損傷與修復研究室
中樞神經損傷是導致成年人永久殘障和死亡的主要原因之一,但至今還沒有有效的治療方法。我們研究團隊 的主要目標是以臨床的角度,運用生理學及分子生物學的技術探討造成頭部外傷及腦中風續發性傷害的病理機轉,並開發可能的治療途徑。
目前,我們利用皮質撞擊腦傷、出血性腦中風、缺血性腦中風動物模式及相關之細胞模式來探討此一問題,應用基因剔除鼠、初代神經元及膠細胞培養、蛋白質分析 (Western blot 及 ELISA)、免疫染色、real-time PCR及動物行為測試等技術。我們從藥理和基因這兩方面着手,主要研究方向為:
1. 以基因剔除鼠來研究caveolin-1、epoxide hydrolase 2 (EPHX2) 、transient receptor potential vanilloid 1(TRPV1) 等不同基因對腦損傷後發炎反應、細胞凋亡和促存活訊號的影響,與相關蛋白質的調節及訊息傳遞。
2. 研究神經保護藥物對小鼠腦傷及腦中風的療效,並進行細胞分子機制探討。
3. 尋找具有鑑別性的生物標誌,協助判斷腦損傷的預後及評估藥物療效。
